چکیده

originated with Gaviola 1 who termed his lifetime instrument a “fluorometer". Clearly the starting point for any fluorescence observation is the light source. Fortunately, we no longer need to rely on sunlight as the principle excitation source for fluorescence. A comprehensive discussion of the development of light sources is beyond the scope of this chapter, but a good index of a wide range of light sources (and many other relevant topics such as filters, monochromators and photodetectors) can be found in the book by Moore et al. [2].

Fluorescence anisotropy measurements in solution: Methods and reference materials (IUPAC Technical Report)*

After recalling the basic relations relevant to both steady-state and time-resolved fluorescence polarization, it is shown how the values of steady-state polarized intensities recorded experimentally usually need to be corrected for systematic effects and errors, caused by instrumentation and sample properties. A list of selected reference values of steady-state fluorescence anisotropy and pola...

Single-molecule FRET of protein structure and dynamics - a primer

Single-molecule spectroscopy has developed into a widely used method for probing the structure, dynamics, and mechanisms of biomolecular systems, especially in combination with Förster resonance energy transfer (FRET). In this introductory tutorial, essential concepts and methods will be outlined, from the FRET process and the basic considerations for sample preparation and instrumentation to s...

Frequency-domain fluorescence lifetime optrode system design and instrumentation without a concurrent reference light-emitting diode.

We report the design, development, and implementation of an improved instrumentation approach for frequency-domain fluorescence lifetime (FDFL) optrodic sensing without a concurrent reference LED. FDFL traditionally uses a reference LED, at approximately the same wavelength as the sensor fluorophore emission, to measure phase shifts associated with changes in the fluorescence lifetime of fluoro...

Toward Quantitative “In Vivo Biochemistry” with Fluorescence Fluctuation Spectroscopy

Quantitative description of protein dynamics and interactions in vivo with temporal and spatial resolution is a key step in dissecting molecular mechanisms in cell biology. Fluorescence fluctuation spectroscopy (FFS) has recently emerged as a powerful in vivo tool for assessing molecular concentration and movement and formation of hetero- and homo-oligomeric complexes. This article discusses po...
